The renewed exchange of attacks comes about a month after both countries signed a preliminary agreement intended to de-escalate hostilities, a deal that has since collapsed.
Iran launched retaliatory strikes across the Gulf after accusing the United States of carrying out intensified attacks on civilian infrastructure, including an airport, railway station and several bridges.
Authorities in Kuwait said an Iranian strike hit an oil facility as well as a power and water plant, while Bahrain's military reported that its air defence systems intercepted multiple Iranian missiles and drones targeting the country.
Tehran also carried out fresh attacks in Jordan, where the United States Central Command confirmed that two American service members were killed on Friday while defending against Iranian ballistic missile and drone attacks. Another service member remains missing.
In a statement broadcast on Iranian state television, Khamenei said the latest attacks demonstrated "the worthlessness of the American president's signature."
"Now that the American enemy seeks to incite war and bear its most serious consequences, it should know that the dear Iranian nation and the axis of resistance have unforgettable lessons to offer it," he said.
Major General Mohsen Rezaei, a senior military adviser to the Supreme Leader, warned that Iran would launch "full-scale offensive operations" if US strikes continued.
According to Iranian state media, Rezaei said Tehran would no longer restrict itself to proportional retaliation, signalling a possible escalation of the conflict.
Kuwaiti authorities condemned the attacks on civilian infrastructure, while residents expressed concerns about potential disruptions to essential services and supply chains.
In Bahrain, the Iranian military claimed responsibility for targeting an air base used by US forces. Iranian state media also reported strikes on fuel storage facilities at Jordan's Al-Azraq military base, following earlier claims by the Islamic Revolutionary Guard Corps that it had attacked US aircraft stationed there with missiles and drones.
Jordan's military said it intercepted 10 missiles on Saturday after shooting down at least three the previous day.
Efforts to revive diplomatic negotiations have largely stalled despite mediation attempts by international actors. US President Donald Trump recently warned that Washington could target additional Iranian infrastructure, although US officials have not confirmed any new operations.
Iran's Deputy Foreign Minister, Kazem Gharibabadi, accused Washington of abandoning its commitments under the previous memorandum of understanding.
"America has violated and stopped all its commitments under that memorandum of understanding, and we have also stopped all our commitments," he said in comments carried by Iranian state television.
Iranian state news agency IRNA reported that recent US strikes killed three people and injured eight in Hormozgan Province. In neighbouring Khuzestan Province, local officials said 95 locations across 12 cities had been targeted over the past 10 days, leaving at least eight people dead, according to Tasnim News Agency.
